چکیده

Recently, an HLA-B*3501-restricted cytotoxic T-lymphocytes (CTL) epitope in the nucleoprotein (NP418–426) of influenza A viruses was identified, which exhibited a high degree of variability. In the present study, the recognition of epitope variants by human cytotoxic Tlymphocytes (CTL) was investigated. Human CD8+ CTL clones were specific for NP418–426 epitope variants within one subtype of influenza virus or cross-reactive with hetero-subtypic variants. Positions in the 9-mer were identified as determinants of CTL-specificity. The in vivo existence of T cells cross-reactive with homoand hetero-subtypic variants of the epitope was confirmed with virusspecific polyclonal T cell populations obtained from HLA-B*3501+ blood donors. Based on CTL recognition patterns and the history of infection as assessed by serology, it was hypothesized that consecutive infections with influenza viruses containing different variants of the epitope select for cross-reactive T cells in vivo. Recognition of homo- and heterosubtypic variants of influenza A viruses by human CD8+ T lymphocytes.

In the present study, the recognition of epitope variants of influenza A viruses by human CTL was investigated. To this end, human CD8(+) CTL clones, specific for natural variants of the HLA-B*3501-restricted epitope in the nucleoprotein (NP(418-426)), were generated. As determined in (51)Cr release assays and by flow cytometry with HLA-B*3501-peptide tetrameric complexes, CTL clones were found...

T-cell tolerance for variability in an HLA class I-presented influenza A virus epitope.

To escape immune recognition, viruses acquire amino acid substitutions in class I human leukocyte antigen (HLA)-presented cytotoxic T-lymphocyte (CTL) epitopes. Such viral escape mutations may (i) prevent peptide processing, (ii) diminish class I HLA binding, or (iii) alter T-cell recognition. Because residues 418 to 426 of the hypervariable influenza A virus nucleoprotein (NP(418-426)) epitope...

Molecular Identification of Pre-Existing Immunityin Human against H9N2 Influenza Viruses Using HLA-A*0201 Binding Peptides

Background and Aims: The contribution genetic and antigenic diversity of H9N2 influenza viruses in evading from immune responses, cytotoxic T lymphocytes (CTL) epitopes in hemagglutinin (HA) protein restricted by HLA binding peptides was identified. Materials and Methods: Phylogenetic analyses were carried out for all of full length HA and deduced amino acid sequences of H9N2 viruses available ...

Sequence variation in a newly identified HLA-B35-restricted epitope in the influenza A virus nucleoprotein associated with escape from cytotoxic T lymphocytes.

Here, we describe a new HLA-B*3501-restricted cytotoxic T lymphocyte (CTL) epitope in the influenza A virus (H3N2) nucleoprotein, which was found to exhibit a high degree of variation at nonanchor residues. The influenza virus variants emerged in chronological order, and CTLs directed against old variants failed to recognize more recent strains of influenza A virus, indicating an escape from CT...

Cross-Protective Peptide Vaccine against Influenza A Viruses Developed in HLA-A*2402 Human Immunity Model

BACKGROUND The virus-specific cytotoxic T lymphocyte (CTL) induction is an important target for the development of a broadly protective human influenza vaccine, since most CTL epitopes are found on internal viral proteins and relatively conserved. In this study, the possibility of developing a strain/subtype-independent human influenza vaccine was explored by taking a bioinformatics approach to...
